Abstract:
using L-sorbose as carbon source in the culture medium, 1, 670 strains of L-sorbose-utilizing bacteria were isolated from 615 samples, an excellent strain N 1197A, from which L-sorbose produces a precursor of vitamin C, 2-keto-l-gulonic acid, was selected. It was later confirmed that the strain was a mixed strain including both large and small colony strains. The two kinds of bacteria were identified. According to The Bergey manual, the large colony strain is Pseudomonas stripe (Pseudo-monas striata); The small colony strain is Gluconobacter oxydans. The fermentation broth was treated with H ~ Type 732 strong acid cation exchange resin, and then concentrated under reduced pressure to obtain white crystals. Through the identification of paper chromatography, elemental analysis and infrared absorption spectrum of vitamin C after conversion, it was proved that the product of N 1197A strain fermented from L-sorbose was indeed 2-keto-l-gulonic acid.

Abstract:
in order to improve the production efficiency of 2-keto-l-gulonic acid (2-KLG) from ketogulgare and Bacillus megaterium (Bacillus megaterium), the effect of pH on K was analyzed. vulgare and B. Effects of megaterium on growth and acid production were found in K.vulgare and B. The optimal growth pH of megaterium was 6.0 and 8.0, respectively. The sugar acid conversion activity of vulgare reaches the maximum value at pH 7.0, so a three-stage pH control strategy (the first stage: 0~8H, pH 8.0; The second stage: 8~20h,pH 6.0; the third stage: 20H to the end of fermentation, pH 7.0) to promote K.vulgare growth and 2-KLG production. The results showed that the implementation of the three-stage pH control strategy further increased the yield of 2-KLG (77.3g/L) and the production intensity (1.38g/(L · h)). And L-sorbose consumption rate (1.42g/(L · h)) increased by 9.7%,33.2% and 25.7%, respectively, compared with the constant pH 7.0.
